Semaglutide units converter (mg to U-100 units)
At a concentration of 2.5 mg/mL, 0.25 mg of semaglutide is 0.1 mL, which is 10 units on a U-100 syringe.

Semaglutide label steps expressed in syringe units
The FDA-approved product is a pen or single-dose device dosed in mg, so this conversion only applies when a vial is drawn with a U-100 syringe. Each mg step below is copied from the Semaglutide 2.4 mg schedule; the units are arithmetic at three example concentrations.
| Step | mg | at 1 mg/mL | at 2.5 mg/mL | at 5 mg/mL |
|---|---|---|---|---|
| Weeks 1–4 | 0.25 | 25 u | 10 u | 5 u |
| Weeks 5–8 | 0.5 | 50 u | 20 u | 10 u |
| Weeks 9–12 | 1 | 100 u | 40 u | 20 u |
| Weeks 13–16 | 1.7 | over 1 mL | 68 u | 34 u |
| Week 17 onward | 2.4 | over 1 mL | 96 u | 48 u |
| Wegovy HD (optional) | 7.2 | over 1 mL | over 1 mL | over 1 mL |
U-100 syringe sizes
Every U-100 insulin syringe uses the same scale, 100 units per mL, so one unit is always 0.01 mL; only the barrel capacity differs. Choose a barrel that holds the full volume, because splitting a draw doubles the measuring error.
| Barrel | Units | Max at 2.5 mg/mL |
|---|---|---|
| 0.3 mL | 30 | 0.75 mg |
| 0.5 mL | 50 | 1.25 mg |
| 1 mL | 100 | 2.5 mg |
Micrograms: 1 mg = 1,000 mcg, so at 2.5 mg/mL each unit holds 25 mcg. Syringe units are not International Units (IU): IU measure biological activity and are defined per drug, as on hCG or somatropin labels.
Worked example
Example: 0.5 mg at 2.5 mg/mL → 0.5 ÷ 2.5 = 0.2 mL → 0.2 × 100 = 20 units.
How it works
1. Find the concentration
Read the concentration in mg/mL (from your own reconstitution arithmetic or the product documentation).
2. Convert mass to volume
Divide the mass in mg by the concentration in mg/mL to get milliliters.
3. Convert volume to units
Multiply milliliters by 100: a U-100 syringe has 100 units per mL.
Frequently asked questions
How many units is 1 mg?
It depends on concentration. Units = (mg ÷ concentration in mg/mL) × 100 on a U-100 syringe. At 5 mg/mL, 1 mg = 20 units; at 2 mg/mL, 1 mg = 50 units.
What is a U-100 syringe?
A syringe marked so that 100 units equal 1 mL; each unit is 0.01 mL.
Are units the same as IU?
No. Syringe "units" are volume markings (0.01 mL each on U-100). International Units (IU) measure biological activity and are drug-specific.
Does this calculator recommend a dose?
No. It converts numbers you enter. Dosing decisions belong to a licensed clinician.
Is Semaglutide measured in units?
FDA-approved Semaglutide pens are dosed in mg. Syringe units only apply when a vial (compounded or research product) is drawn with a U-100 syringe, and depend on its concentration.
